Can Trailer Wheel Pilot Diameter be Larger than Needed and Work

Does the pilot hole have to be the same size as the hub or can it be bigger

As long as your trailer hub has studs for attaching the trailer wheel and not threaded holes for wheel bolts all you need in regards to wheel pilot diameter is one that is large enough to fit on the hub. So bigger is totally fine.
The part # AM35351DX that you referenced has a pilot diameter of 2.84 inches.
